You have to pick Brian up and take him to the place where the deal is taking place and then take him back.
You have to meet Brian in Downtown Broker, take him near Asparagus Avenue, and then save him from the drug dealers.
Random characters are minor characters you can find around Liberty City. They provide you short missions, most of which are necessary for 100% completion of the game.
